The 1991 paper with Axel showed that hundreds of genes produce odor sensors on neurons in the nose. Each sensor is a protein that changes shape when an odor binds to it, sending an electrical signal to the brain. Because different sensors respond to different odors, the brain interprets these signals as specific scents. Buck and Axel cloned these receptors, showing they belong to the G protein-coupled receptor family. By analyzing rat DNA, they estimated about 1,000 different genes for olfactory receptors in mammals. This work opened up the genetic and molecular study of smell. Later, they showed that each olfactory neuron expresses only one type of receptor, and all neurons with the same receptor send their signals to a single dedicated spot in the olfactory bulb.

She was elected to the National Academy of Sciences in 2003 and the Institute of Medicine in 2006. She has been a Fellow of the American Association for the Advancement of Science and the American Academy of Arts and Sciences since 2008. She also serves on the Selection Committee for Life Science and Medicine for the Shaw Prize. In 2015, Harvard University gave her an honorary doctorate, and she was elected a Foreign Member of the Royal Society.
